Is the minimum unit for the coordinates and translation distance of nodes in glyphs 1. What do I need to do if I want to assign coordinates to nodes (100.5,100.5) instead of (101,101)?

You can define a finer grid in Font Info > Other. Set the subdivision to 2. Or 10 if you need 10.1.
Those coordinates can be exported to OTF, but not to TrueType and Variable fonts. If that is an issue, you can increase the UPM (units per em) in Font Info > Font. Use the arrow button to rescale the whole font to 2000 or 4000 units.
