Couldn’t find the exact match to my problem, so here goes;
Stylistic set #1 contains a ‘schoolbook’ a, but it also need to exist as a rounded feature(accents etc), which is stylistic set #2, but that contains the regular ‘a’. How to do?
This is rather cryptically worded… could you clarify what exactly you mean? SS01 also needs to exist as SS02, but a.ss01 should use a different a glyph than a.ss02?

So you have a stylistic set for single-storey a (SS01) and a stylistic set for rounded dots (SS02); you would indeed combine them as Sebastian wrote. The glyphs in your screenshot would then be:
a a.ss01 adieresis.ss01 adieresis.ss02 adieresis.ss01.ss02
This is assuming, of course, that the base adieresis has the two-storey a and square dots.
