I am very new to the program, I need to update one of the alphabet and after I pasted the new one in, the colour of the number change (see screenshot) and nothing move (the alphabet) when I change the number. Appreciate if anyone can give me some guidance.

When using arithmetics, use a = prefix, so =n-100, for example.

Two things: You are doing spacing, not kerning. For details read this: Spacing | Glyphs and Kerning | Glyphs
And please don’t overuse metrics keys. Only use them for shapes that are very similar (e.g. the left side of the “b” and “h”, or “n” and “m”).

Metrics keys are the formulas you put into the side bearing fields (“=n-100”).
